ABSTRACT

【Objective】 To establish an indirect ELISA method for detecting IgG, IgM and IgA antibodies against Hepatitis E virus (HEV). 【Methods】 Tn-5 cells were infected with recombinant HEV baculovirus, and HEV-like particles (VLP) were collected and purified for coating antigen. The reaction conditions and methodology of indirect ELISA method were established and evaluated. The prevalence of HEV antibody among blood donors in Xishuangbanna were detected. 【Results】 The collected and purified VLP showed HEV antigenicity. The positive rates of anti-HEV IgG, IgM and IgA in blood donors in Xishuangbanna Prefecture were 18%(90/500), 5.6%(28/500) and 2.6%(13/500), respectively. The recent HEV infection rate was 1.8 % (9/500), and the seroprevalence of hepatitis E was 19.8% (99/500). Of the 13 anti-HEV IgA positive samples, 2 were both anti-HEV IgG and IgM negative. 【Conclusion】 HEV antibody positive is common among blood donors in Xishuangbanna, some of which are recent infections, posing a threat to the safety of blood transfusion. HEV IgA antibody indirect ELISA combined with human HEV IgM antibody detection can improve the detection rate of recent HEV infection.

ABSTRACT

Periodontitis patients are at risk of alveolar bone loss during orthodontic treatment. The aim of this study was to investigate whether intermittent parathyroid hormone (1-34) treatment (iPTH) could reduce alveolar bone loss during orthodontic tooth movement (OTM) in individuals with periodontitis and the underlying mechanism. A rat model of OTM in the context of periodontitis was established and alveolar bone loss was observed. The control, iPTH and iPTH + stattic groups received injections of vehicle, PTH and vehicle, or PTH and the signal transducer and activator of transcription 3 (STAT3) inhibitor stattic, respectively. iPTH prevented alveolar bone loss by enhancing osteogenesis and suppressing bone resorption in the alveolar bone during OTM in rats with periodontitis. This effect of iPTH was along with STAT3 activation and reduced by a local injection of stattic. iPTH promoted osteoblastic differentiation and might further regulate the Wnt/β-catenin pathway in a STAT3-dependent manner. The findings of this study suggest that iPTH might reduce alveolar bone loss during OTM in rats with periodontitis through STAT3/β-catenin crosstalk.

ABSTRACT

OBJECTIVETo investigate the replacement therapy of rPTH(1-84) (recombinant human parathyroid hormone (1-84)) to hypothyroidism in established rat model.

METHODRat model of hypothyroidism was established by resecting parathyroids. A total of 30 rats with removal of parathyroids were divided into 6 groups randomly, 5 in each group, and applied respectively with saline injection (negative control group), calcitriol treatment (positive control group) and quadripartite PTH administration with dose of 20, 40, 80 and 160 µg/kg (experimental groups). Saline and rPTH(1-84) were injected subcutaneously daily. Calcitriol was gavaged once a day. Sham-operation was conducted in 5 rats of negative control group. To verify the authenticity of the rat model with hypothyroidism, the serum was insolated centrifugally from rat blood that was obtained from angular vein at specific time to measure calcium and phosphorus concentration. Urine in 12 hours was collected by metabolic cages and the calcium concentration was measured. After 10-week drug treatment, the experiment was terminated and bilateral femoral bone and L2-5 lumbar vertebra were removed from rats. Bone mineral density (BMD)of bilateral femoral bone and lumbar vertebra was analyzed by dual X-ray absorptiometry (DXA). The concentration of bone alkaline phosphatase (BALP) in serum was determined by radioimmunoassay.

RESULTThe rat model with hypothyroidism was obtained by excising parathyroid gland and was verified by monitoring calcium and phosphorus concentration subsequently. Administration of rPTH(1-84) in the dose of 80 or 160 µg/kg made serum calcium and phosphorus back to normal levels, with no significant difference between the doses (P>0.05). The BMD in each group of rats with rPTH(1-84) administration was increased significantly (P<0.05). The levels of urinary calcium and serum BALP in rats of maximum rPTH(1-84) injection group (160 µg/kg) were higher than those of normal control group (P<0.05). The rats treated with calcitriol had normal calcium levels and showed the increase of BMD and phosphorus concentration compared with normal control group (P<0.05). The amount of urinary calcium also exceeded the other groups (P<0.05), but no with significant difference in BMD of bilateral femoral bone and lumbar vertebra between negative control group and normal control group (P>0.05).

CONCLUSIONCalcium and phosphorus return to normal level by administration of rPTH(1-84) in the dose of 80 µg/kg or 160 µg/kg, with increase in BMD. Calcitriol can return the level of calcium to normal and increase BMD, but can not correspondingly decrease the phosphorus concentration and increase the excretion of calcium in urine.

ABSTRACT

OBJECTIVE@#To determine the most appropriate form of selective neck dissection(SND) in the hypopharyngeal cancer with cervical lymph node metastasis.@*METHOD@#We have retrospectively analyzed the distribution and prevalence of cervical metastasis in 26 patients with hypopharyngeal squamous cell carcinoma from January 1998 to December 2008. All the patients underwent SND as part of the primary treatment. There were 34 elective SNDs and 17 therapeutic SNDs from 11 node-negative hypopharyngeal cancers and others node-positive.@*RESULT@#Occult metastasis was found in 6 patients (55%) with cervical metastasis confined to level II and III. Clinical node-positive necks were all pathologically identified with 6.7%, 66.7%, 86.7%, 46.7%, and 20.0% of the prevalence of metastasis to level I, II, III, IV and V respectively. The regional recurrences were found in 4 patients during the follow-up, which were all from cN+ patients. No patient experienced level I recurrence.@*CONCLUSION@#The results of this study suggest that SND (I-III) may be feasible for the treatment of cN0 hypopharyngeal cancer, which needs a larger sample to verify. Meanwhile, from our data, it has a satisfactory result to perform SND (II-V) with adjuvant radiotherapy for the cN+ patients.

ABSTRACT

Co-fermentation of glucose and xylose is critical for cellulosic ethanol, as xylose is the second most abundant sugar in lignocellulosic hydrolysate. In this study, a xylose-utilizing recombinant Zymomonas mobilis TSH01 was constructed by gene cloning, and ethanol fermentation of the recombinant was evaluated under batch fermentation conditions with a fermentation time of 72 h. When the medium containing 8% glucose or xylose, was tested, all glucose and 98.9% xylose were consumed, with 87.8% and 78.3% ethanol yield, respectively. Furthermore, the medium containing glucose and xylose, each at a concentration of 8%, was tested, and 98.5% and 97.4% of glucose and xylose was fermented, with an ethanol yield of 94.9%. As for the hydrolysate of corn stover containing 3.2% glucose and 3.5% xylose, all glucose and 92.3% xylose were consumed, with an ethanol yield of 91.5%. In addition, monopotassium phosphate can facilitate the consumption of xylose and enhance ethanol yield.

ABSTRACT

A robust strain of the species Saccharomyces cerevisiae CGMCC1949 was screened and identified, and advanced solid state fermentation (ASSF) technology for fuel ethanol production from sweet sorghum stalks was thus developed. The fermentation time was shortened to less than 30 h, and ethanol yield was 92% of its theoretical maximum. And in the meantime, the cost-effective storage was established for sweet sorghum stalks, with less than 5% sugar loss after 200 days of storage, making the plant operation could extend up to 200 days without feedstock shortage. With the fermentation kinetics and heat-mass transfer models, modeling of the ASSF process was investigated, and the rotating drum bioreactor was designed. Furthermore, the ASSF technology was successfully applied in the pilot plant in which the rotating drum bioreactor was scaled up to 127 m3, and ethanol yield of 91% was achieved. At the end, techno-economic analysis (TEA) conducted by ASPEN indicated that ethanol production from sweet sorghum stalks by the ASSF is economically competitive.
